Title / Titel Effects of d-amphetamine on sensorimotor gating, decision making and psychological measures. A double-blind placebo-controlled dose-effect study in man.

Summary / Zusammenfassung Animal studies demonstrate that increased dopaminergic activity in limbic brain regions disrupts sensorimotor gating as indexed by prepulse inhibition (PPI) of the startle reflex. Gating deficits may render schizophrenia patients to attentional deficits and psychosis. However, whether increased dopamine activity results in PPI deficits in man, and how such PPI deficits are linked to cognitive abnormalities has not yet been established humans. The present study aims to further elucidate the effect of d-amphetamine, an indirect dopamine agonist, on PPI and cognitive performance in healthy human subjects (n=16). The plasticity of sensory gating will be assessed by measuring prepulse inhibition and habituation of the acoustic startle reflex. Cognitive performance (attention, memory, planning and strategy) will be assessed using neuropsychological tasks from the Cambridge Neuropsychological Test Automated Battery (CANTAB), sequential behavior in decision making investigated using a computerized two-choice prediction task (Paulus et al, 2000). Subjective effects of psilocybin on mood states are assessed by use of the EWL-60 mood rating scale, and dose-dependent alterations in consciousness are rated via 5-D ASC questionnaires.
